View SamplesWe discovered lncRNA PSTAR as a novel hepatocellular carcinoma (HCC) - associated lncRNA, which could interact with hnRNP K protein. To explore the function of PSTAR in HCC and underlying molecular mechanism, we examined the global gene expression change in HepG2 cells with PSTAR knockdown. The gene expression profile of HepG2 with hnRNP K knockdown was also analyzed to investigate the correlationship between PSTAR and hnRNP K. Further, doxorubicin (DOX) was applied to induce DNA damage, and the effect of PSTAR and hnRNP K under DNA damage was also investigated.

View SamplesWe integrated the copy number data with gene expression data from the same HCC samples, and identified fifteen putative driver genes with recurrently genomic aberrations and their associated modules in HCC. We further confirmed empirically that three putative driver genes (MYH10, CEP104 and RRS1) play significant roles in tumor initiation and progression of HCC. Notably, we demonstrated that RRS1 regulates the MDM2-P53 pathway and promotes tumor progression by retaining RPL11 in the nucleolus in HCC. Altogether, these data provide insights into novel cancer driver genes and suggested molecular targets for treatment for HCC.

View SamplesIn space, multiple unique environmental factors, particularly microgravity and space radiation, pose constant threat to the astronaut’s health. To gain insight into the role of miRNAs and lncRNAs in response to radiation and microgravity, we analyzed RNA expression profiles in human lymphoblastoid TK6 cells incubated for 24 h in static condition or in rotating condition to stimulate microgravity in space after 2 Gy γ-ray irradiation. Expression of 14 lncRNAs and 17 mRNAs was found to be significantly down-regulated in the simulated microgravity condition. In contrast, irradiation up-regulated the expression of 55 lncRNAs and 56 mRNAs, while only one lncRNA, but no mRNA, was down-regulated. Furthermore, 2 miRNAs, 70 lncRNAs, and 87 mRNAs showed significantly altered expression under simulated microgravity after irradiation, and these changes were independently induced by irradiation and simulated microgravity. Together, our results indicate that simulated microgravity and irradiation additively and independently alter the expression of RNAs and their target genes in human lymphoblastoid cells.
