Total white cells were purified from peripheral blood of BCR/ABL+ CML patients by Ficoll-Hypague (Amersham Pharmacia Biotech) density gradient centrifugation. CD34+ cells were positively selected using the Midi-MACS immunomagnetic separation system (Miltenyi Biotec, Bergisch Gladbach, Germany) as previously described (Steidl et al., 2002). Purities of isolated CD34+ CML cells ranged between 90 and 99%. Total RNA was isolated from CD34+ cells with RNeasy Mini Kit (Qiagen AG, Hilden, Germany) according to the manufacturer's instructions. High quality RNA was assessed using an Agilent 2100 Bioanalyzer (Agilent, Palo Alto, CA, USA). None of the samples showed RNA degradation or contamination by DNA. Samples had been stored at -80 C until use in gene expression analyses.
